How do I draft the email saying all of the above, including my colleague is away and that I’m picking this up for him...

If your colleague wants them to respond to his email, why can't he email them himself?
plain English works fine. the target of the email will either do it or s/he won't -- that isn't within your ability to control. I'd use a heading such as "Person X asks ..." -- assuming that the person you're doing this for carries some weight with the target
