I want to give a Japanese title to something I'm creating. Would this phrase be an acceptable title? I ask this because after google searching the phrase with quotes nothing came up.
The basic meaning I'm trying to convey is something like "bearer of burdens."

No, it doesn't work, though it's grammatically OK. "おもにのしょじ" means "to possess burdens / possession of heavy luggage", which wouldn't be your intention.
"Bearer of burdens" would be translated like this:
重責の担い手(じゅうせきのにないて / juuseki no ninaite)
Literally, 重責(じゅうせき) means "heavy responsibility", and 担い手(にないて) means "bearer / carrier".
yes, it's perfect.
