I forgot to return a vest that was a part of my uniform for a hotel that I quit working at months ago. At the time, my front desk manager said that if he needed it, he would let me know before my last paycheck. Well that was two months ago and now he’s trying to say I have to pay them $100 since I never returned it and no longer have it. However, I have statements from two former employees who also never returned their uniforms and were never ever asked to pay the hotel back.

Yes. What other employees did or did not do does not concern you and what you were told by someone does not relieve you from your obligation. It DID NOT belong to you. It belonged to them and you know it. Pay up, or you will regret it. There is much that they can do to make you suffer if you don't.
You literally CAN'T know they don't do the same thing on a regular basis. Even if they don't, you would have to prove they treated you differently because you are a member of a legally protected class in order to have a case on that basis.
Does he have your name on any sort of paperwork which states you will be charged for such things?
If not, tell him to sue you. He isn't going to bother.
What happens with other people doesn't effect your case at all.
But if what you want is to be a little sht about it, then just don't pay them. Then they'd have to take you to court. The thing is, they'll have to pay someone to file in court. So they won't sue you.
