The first one that flashed into my mind was "Midnight Cowboy." The last scene, in which Joe Buck finally makes it down to Florida, only to discover that his friend "Ratso" has died on the way, is one of the saddest sequences ever filmed. The two of them on that bus in their cheesy Hawaiian shirts...just tears your guts out.
