The idea that the only justification for exercising power over someone is that their behaviour would otherwise cause harm to another. We (or the state, which Mill was concerned with) should not interfere with personal liberty because we do not like what the person is doing; we should only get involved when they are, or are about to, hurt someone other than themselves.
The basic significance is that people should have the liberty to do whatever they please, to themselves and to others, so long as they don't hurt others while doing it.
