The first patent that Samuel Colt pulled, was for the first revolving pistol, the Paterson. It was a cap and ball, 5 shot revolver. It was not 45 caliber, so those that answered 6 would be correct.

The patent acquired in 1836 was for revolvers that would fire 5 or 6 shots without reloading.
