The Hebrew word rendered "blameless" (NKJV) or "perfect" (KJV) in Genesis 17:1 means "entire, complete, full, without blemish." The Greek word found in Matthew 5:48 translated "perfect" means "finished, complete, having reached its end," and implies being fully grown or mature. The definition of the English word perfect is "lacking nothing essential to the whole, without defect, complete." All three definitions contain the word "complete.
There is no contradiction in the Bible concerning this concept. The doctine is called sanctification -- and claims that God's people go through a process which leads to an end result in eternity where they take on various characteristics of Christ including holiness and perfection/completeness. (Someone has already posted the explanation of the Greek word involved in these passages.) The fact that people do not immediately assume the full measure of sanctification does not contradict what the Bible claims on this subject.
But I'm impressed that so many people who do not presently understand these concepts are asking questions and learning about the scriptures as to these topics. There seems to be a great deal of interest in the Biblical text here. No wonder the Bible remains the most published book of all time and the perpetual best seller. Indeed, even those who hate the Bible tend to speak of it often. That has always fascinated me -- especially considering that I used to be one of them (and I ended up studying both of the primary Biblical languages in an effort to debunk the Biblical texts.)
It is Gods command that we become perfect and sinless in our inner selves by believing in His gospel of the water and the Spirit (John 3:5).
The reason for this is because no sinner may enter the kingdom of heaven. So it is here on this earth that we need to become righteous. And we must not read the Bible literally like many do and suffer.

God gives the gift of His own love through the Spirit......
"And hope does not disappoint us, because God has poured out his love into our hearts by the Holy Spirit, whom he has given us. "

The Spirit is received by faith in the message of the Gospel:
"that God was reconciling the world to himself in Christ, not counting men's sins against them. And he has committed to us the message of reconciliation. We are therefore Christ's ambassadors, as though God were making his appeal through us. We implore you on Christ's behalf: Be reconciled to God. God made him who had no sin to be sin for us, so that in him we might become the righteousness of God."
Since the bible says no man is righteous, why even ask man to be perfect?
Obviously, Jesus already knows man cannot do it, but yet are to continue working towards it. Maybe that's why Paul said 'to strive towards the upward call' and to work out the salvation with fear and trembling.
All adds together to mean 1 thing. Once saved is not always saved.
God made the rules and realized that humans cannot follow all of them to perfection, that is why he gave his son Jesus Christ to die for our imperfections(sin) so that any time we fail to follow his rules we can ask for forgiveness in the name of Jesus and we will be forgiven...
So that we would come to Him to be saved in Christ Jesus,our failings be forgiven through His death in our place, and the Holy Spirit of God bring us to new birth as sons and daughters of God. And in this new life in Christ, learn to live by grace and the law of love.

I am to press onwards with Christ to become daily more like God -
all the while knowing that perfection comes when I am taken from here and translated into the Kingdom of Heaven.
This is part of the beauty of God; that everything He commands of His children, He wilfully gives.
All that He asks He provides.

If god is perfect and created humans in his image you would expect humans to be perfect as well. I have yet to hear about one perfect human being....
