So since I work as a cashier, and say a customer comes in and I can’t tell if they are male or Female?? Would that be OK to ask them if they are a Man or a Woman? Or would they think I’m being a rude jerk?

Yes, I suppose you need to get used to removing gender from the language (e.g. not saying "sir" or "ma'am" might take some getting used to), but it really doesn't effect the sales transaction, so I ask again ...

As a cashier your job is to take their money and give them a receipt. There's absolutely no reason their gender should be important to you. In fact outside of medical professions I can't think of many realms of employment where the gender of a customer would matter. Just do your job and don't worry about what "Pat" is doing.
Their name should give you a hint.
